Malaria and leishmaniasis are diseases with a significant socioeconomic impact, exacerbated by the resistance and toxicity of available treatments, which drives the search for new therapeutic alternatives. Traditional communities use Brazil nut (Bertholletia excelsa) tea for liver treatment and as an antimalarial agent, constituting a valuable starting point for scientific investigations into its bioactive compounds. The objective of this study was to evaluate the susceptibility of Plasmodium falciparum and Leishmania amazonensis to the action of metabolites present in crude extracts of different plant structures of B. excelsa. Based on the results obtained, a scientific article was prepared, structured according to the institutional norms for academic work of this type. Samples consisting of stem bark, fruit, and operculum (the structure inside the fruit) were collected from native Amazonian trees in Porto Velho/RO. The plant samples, dried in an oven, had their metabolites extracted using water at room temperature (25°C), water at 100°C, and methanol. Once dried, the crude extracts were analyzed by ultra-high performance liquid chromatography coupled to high-resolution tandem mass spectrometry (UHPLC/HRMSn) using a C18 column and electrospray ionization in positive (ESI+) and negative (ESI-) modes. Analyte identification was performed using MS Dial 5.1 software. The results reveal an abundance of secondary metabolites, described for the first time in B. excelsa, belonging to the classes of flavonoids, coumarins, and terpenes (triterpenes, diterpenes, sesquiterpenes, and monoterpenes), expanding knowledge about the diversity and chemical richness of this species. The identified metabolites, aligned with the biological activities described in the literature, confirm the biological potential of this species. Biological assays demonstrated significant antioxidant activity against the DPPH free radical, with minimum IC50 values of 1.48 µg/mL for the stem bark extract, 14.15 µg/mL for the fruit extract, and 13.25 µg/mL for the operculum extract. Additionally, they exhibited potent inhibitory activity against chloroquine-resistant P. falciparum, with inhibitions between 80 and 100% at a concentration of 100 µg/mL. The minimum IC50 values for this antiplasmodial activity were 2.38 µg/mL for the stem bark extract, 9.41 µg/mL for the fruit extract, and 30.36 µg/mL for the operculum extract, scientifically validating the traditional use of Brazil nut. It was also found that the crude extracts of stem bark and fruit showed greater selectivity for the parasite than for the HePG2 and VERO cell lines, justifying further studies. However, no relevant minimum inhibitory activities were observed against the tested L. amazonensis species. These results highlight the importance of ethnobotanical knowledge as a guide for bioprospecting, demonstrate the potential and importance of Amazonian biodiversity as a source of new bioactive compounds, and reinforce the need for the preservation of plant species in the region.

Currently, research is being conducted to create new-generation insecticides by utilizing molecules that interact with specific molecular targets, such as enzymes linked to trehalose metabolism and other targets related to essential insect functions. Thus, the objective of this work was to model, perform virtual screening, and molecular docking of the Trehalose 6-Phosphate Phosphatase enzyme from Aedes aegypti (AaeTPP) with Phthalimide derivatives and the soluble resin fraction (SF) of Bertholletia excelsa (Brazil nut tree), and to evaluate the larvicidal effects of one of the Phthalimide derivatives, i.e., N-(p-tolylsulfonyl)phthalimide (PNT), and the soluble resin fraction of B. excelsa against Aedes aegypti.
The three-dimensional structure of the AaeTPP enzyme was modeled by homology. Following structural validation, virtual screening of PubChem compounds was performed based on similarity to the substrate Trehalose-6-Phosphate (T6P) and the inhibitor N-(phenylthio)phthalimide (NPP), as well as to the metabolites present in the soluble resin fraction of B. excelsa. These compounds were then subjected to molecular dynamics simulation, and the analyses indicated stability and highlighted ligands with inhibitory potential.
A stock solution of the PNT molecule was prepared with 145 mg of PNT in 50% dimethyl sulfoxide (DMSO) and 50% deionized water, and concentrations of 100, 50, 25, 10, and 5 ppm were tested. Larvicidal tests were carried out in a 24-well Elisa plate in triplicate with 5 larvae (L1) with a final volume of 1 mL and a control with 0.5% DMSO. A stock solution was prepared with 5 g of B. excelsa resin 400.1 and 208.3 in 95% Methanol and 5% DMSO, and the SF was tested at concentrations of 224, 560, 784, 1120, 1680, and 2240 ppm. Larvicidal tests were performed in 50 mL plastic containers with a final volume of 10 mL, in triplicate with 10 third-instar larvae, and the control with 1% of the solution used to dilute the resin in distilled water. Mortality in the experiments was recorded at intervals of 24–96 hours.
The modeling of the AaeTPP enzyme was based on the structure of TPP from Mycobacterium tuberculosis (PDB: 5GVX), showing 30.8% similarity. The structure had an RMSD of 0.603 A˚ and 80.23% positive validation by Verify3D. 154 Phthalimide derivatives exhibited a better binding energy than the natural substrate T6P (−5.18 kcal/mol), and 50 compounds from the B. excelsa soluble fraction showed favorable energy (−9.75 kcal/mol). In the molecular dynamics simulations (150 ns), the AaeTPP–PNT complex showed greater structural stability than T6P. The analysis indicated that PNT had the most favorable binding energy (−16.68 kcal/mol).
PNT concentrations between 10 and 100 ppm caused 38% to 52% mortality after 24 hours of exposure, reaching 92% at 100 ppm after 72 hours. We observed that, in both B. excelsa samples, concentrations from 224 to 1120 ppm did not cause larval mortality. The Estimated Lethal Concentrations (LC) of PNT required to kill 50% (LC50), although tending to decrease over the observation time, did not significantly differ from each other. The LC90 values, however, were 7 times higher than the LC50 at 24 hours, reaching 11 times higher after 48 hours. The LC50 values for both resin samples decreased over time: for sample 400.1, LC50 was 1741 ppm (48h), 1411 ppm (72h), and 1117 ppm (96h). For sample 208.3, LC50 was 1816 ppm (48h), 1562 ppm (72h), and 1493 ppm (96h).
The increase in concentration and exposure time of Ae. aegypti larvae to N-(p-tolylsulfonyl)phthalimide demonstrated larvicidal potential, whereas the resin fractions of B. excelsa showed low larvicidal potential against Ae. aegypti immatures.

The Iquiri National Forest (FLONA do Iquiri), located in the southwest of the Brazilian Amazon, is home to traditional communities whose livelihood is deeply connected to the sustainable management of natural resources. This study aimed to conduct a socio-economic and environmental assessment of these communities, with an emphasis on agro-extractive activities, in order to understand their productive dynamics, identify challenges, and propose strategies to strengthen social production while promoting environmental conservation. The research adopted a transdisciplinary and participatory approach, applying 24 semi-structured interviews with farmers and extractivists living in the unit. Data were analyzed through descriptive statistics and content analysis. Six agricultural products were identified (with cassava present in 100% of the families) and five plant-based extractive products, such as Brazil nuts (collected by all 24 families), copaiba oil (produced by 21 families), sorva, vine, and acai. The average annual family income from extractive activities was estimated at R$ 11,166.91, with Brazil nuts and copaiba standing out. In agriculture, cassava generated an average annual income of R$ 3,747.92 per family. The data also revealed the central role of women in agricultural and extractive management and the strong dependence of communities on the seasonality of natural resources. The feasibility analysis showed that, despite productive potential, the communities face limitations related to infrastructure, precarious river transport, lack of local processing, and dependence on middlemen, which reduces the added value of the products. Comparative experiences with other Conservation Units, such as FLONA do Tapajos, demonstrated that the adoption of integrated public policies, combined with community organization and technical support, can significantly increase the economic sustainability of agro-extractive activities. It is concluded that FLONA do Iquiri presents high potential to consolidate sustainable management practices that reconcile forest conservation, strengthening of family-base economy, and appreciation of traditional knowledge of local populations, serving as a model for socio-environmental development in the Amazon. Aquaculture in Brazil has established itself as an important activity within Brazilian agribusiness, and in 2026, national production of farmed fish reached 1,011,540 tons, with tambaqui (Colossoma macropomum) standing out as the main native species produced in the country, accounting for 257,070 tons, placing the state of Rondônia in a prominent position as the leading producer of the species, responsible for 55,880 tons. However, aquatic health remains an obstacle to production, with various substances being used off-label to control microorganisms, negatively impacting the environment where they are produced and, consequently, posing risks to human health. Due to this, in search of alternatives, the use of immunomodulators in aquaculture has been studied. Among the plants with this potential is pariparoba (Pothomorphe umbellata), widely used in traditional medicine and with immunostimulant potential. Due to the need for alternatives to the use of chemotherapeutics in fish farming and the potential of pariparoba, the present study sought to evaluate the immunomodulatory activity through the supplementation of pariparoba leaves in tambaqui, assessing hematology, histopathology, and immunohistochemistry in acute inflammation. For this, four concentrations of supplementation using the plant's leaves were tested, added to commercial feed, and provided daily to the animals for thirty days. After this period, the groups were challenged, and blood and liver, spleen, and kidney tissue samples were collected at 12, 24, and 48 hours. Regarding hematimetric indices, there was no significant change in RBC, Ht, MCV, MCHC, and MCH at any of the concentrations tested. In leukocyte activity at 1% supplementation, a lower count was observed at the evaluated times, suggesting an anti-inflammatory effect. In groups 2, 3, and 4, no difference was observed compared to the positive control. In groups 2, 3, and 4, no difference was observed in relation to the positive control (p<0.05). In the analysis of phagocytes, after 48 hours, a reduction in activity was observed in all treatments analyzed, with emphasis on treatments 1, 2, and 3%, suggesting that lower doses of supplementation present better immunomodulation. In the histopathological analysis of the spleen, liver, and posterior kidney, no alterations were observed that compromised organ function. The percentage of immunolabeling of the cytokine IL-10 showed a response dependent on time and supplementation percentage, with greater immunolabeling observed after 24 and 48 hours of inflammatory induction. In iNOS, the liver was the organ that showed the highest labeling at all times (p<0.05), followed by the kidney and spleen. The results indicate that supplementation with pariparoba leaves at the tested concentrations was able to alter leukocyte expression, phagocytic activity, and IL-10 and iNOS expression, without causing changes in hematimetric, erythrocyte, and histological indices. Supplementation with 1 to 2% of pariparoba leaves for thirty days proved to be more effective in regulating the induced inflammatory process, without deleterious tissue alteration.

This dissertation assessed the functional trajectory of the Bom Futuro National Forest, a protected area located in Porto Velho, Rondônia, using biophysical indicators derived from the PySEBAL algorithm and counterfactual scenarios based on Random Forest models. The first chapter analyzed the temporal trajectory of aboveground biomass production (AGBp), biomass deficit (AGBd), and biomass water productivity (WPb) during the dry season from 2013 to 2025, using Landsat 8 imagery. The Mann–Kendall test with autocorrelation correction, Sen’s slope estimator, Pettitt’s change-point test, and the Wilcoxon test with Cohen’s d estimator were applied, segmented across seven edge zones. The results indicated consistent functional decline: AGBp showed a significant negative trend in approximately 30% of the analyzed area, with a median rate of −2.889 kg ha⁻¹ day⁻¹ year⁻¹ and a more intense deterioration gradient in edge zones; WPb exhibited a structural break in 2020 in more than 50% of stable pixels, with no recovery in subsequent years; AGBd confirmed the intensification of water stress after the break. The second chapter employed Random Forest models with spatial block cross-validation to construct counterfactual impact assessment scenarios across seven areas subjected to ecological restoration interventions. The models showed satisfactory performance for biomass and water productivity variables (CV R² between 0.925 and 0.963), and the endogeneity diagnosis, based on mean-shift Z-scores, did not identify violations of the exogeneity assumption in any of the 52 tests performed. Impact estimates were predominantly negative in the first six years post-intervention, interpreted as a reflection of non-stationarity caused by the 2020 regime shift, which compromises the extrapolation of the counterfactual baseline. A positive reversal trend observed in years 7 and 8 post-intervention is consistent with late-stage functional recovery patterns described in the literature. It is concluded that the Bom Futuro National Forest exhibits progressive metabolic deterioration, aggravated by a functional regime shift beginning in 2020, and that the integration of PySEBAL, non-parametric time series analysis, and machine learning-based counterfactual modeling constitutes an operationally viable approach for large-scale ecological restoration monitoring in the Amazon.

Copaiba trees (Copaifera spp.), widely distributed in the Amazon, produce an oleoresin with antioxidant, anti-inflammatory, antimicrobial, and healing properties. This oil is used in the cosmetics, paint, and lubricant industries, and in traditional medicine, especially in the Northern region of Brazil. Despite its potential, the productive arrangement within Extractive Reserves (Resex) in Rondônia remains weakly structured, rendering this bioeconomy largely invisible. This study analyzes the ecological, productive, and market dimensions of copaiba oil extraction and trade by extractivist communities in Rondônia. We interviewed three groups—G1: extractivists, G2: traders, and G3: fostering agencies—across conservation units in the municipalities of Costa Marques, Guajará‑Mirim, Machadinho do Oeste, and Vale do Anari, covering the full chain from tapping to commercialization. The assessment focused on ethnobotanical knowledge, management practices, processing, storage, value‑addition strategies, cultural aspects, and a SWOT analysis (Strengths, Opportunities, Weaknesses, and Threats). Fieldwork was conducted in the Aquariquara, Castanheira, Rio Cautário, Rio Pacaás Novos, and Rio Preto Jacundá Resex. In total, 40 participants took part: G1 (17), G2 (10), and G3 (13). The study also evaluated the oil’s economic contribution to local households and identified opportunities to strengthen the Amazonian bioeconomy through the valorization of traditional knowledge and better production organization. Results indicate that the local productive arrangement of copaiba generates significant supplemental income for extractivists. Four oleoresin types were cited—Mari‑Mari, Angelim, Cuiarana, and Jacaré—with mean first‑tapping yields of approximately 10, 40, and 20 liters, respectively, for the first three types. Sale prices ranged from R$60.00 to R$120.00 per liter, with local sales and shipments to São Paulo, Minas Gerais, Espírito Santo, and Bahia. Recommended good practices include sealing the trunk borehole with a breu‑wood plug, filtering to remove residues, and storing oil in clean, opaque containers. Identified weaknesses include the lack of adequate management, loss of productive areas to agribusiness, and a fragile institutional support network. We conclude that copaiba oil production is viable, but greater community organization and public policies for technical assistance, credit, promotion, appropriate management practices, and traceability are needed. These actions can strengthen the local bioeconomy, improve livelihoods, and add value to standing forests, ultimately enhancing quality of life in the traditional communities studied.

Deforestation in the Brazilian Amazon is a complex phenomenon, whose drivers are widely debated in the scientific literature, but frequently analyzed at aggregate scales that obscure local dynamics. This work investigated the drivers of forest loss through a mixed-methods approach, combining a Systematic Literature Review (SLR) and spatially explicit predictive modeling. The SLR, based on 135 articles, revealed a significant methodological bias: 79% of the drivers identified as relevant are concentrated at macro and meso-scales, generating a knowledge gap about the factors operating at the property level and within protected areas. To mitigate this gap, a case study was conducted in the Conservation Units (CUs) of Rondônia, using the Random Forest machine learning algorithm and the SHAP interpretability technique at a microscale (250 m resolution). The model obtained an average AUC of 0.81 and identified "distance to internal deforestation" as the variable with the greatest predictive power, demonstrating that pre-existing environmental liabilities act as catalysts for new conversions through a contagion effect. The results highlighted extreme institutional vulnerability, with the risk concentrated in state-managed and Sustainable Use protected areas, notably the Jaci-Paraná Extractive Reserve, suggesting that internal degradation precedes and justifies political processes of reduction or declassification of these areas (PADDD). It is concluded that the effective protection of protected areas depends not only on legal barriers, but also on the immediate containment of internal logistical expansion and the strengthening of governance at the state level.

Acalypha arvensis is a plant widely used in traditional medicine and with immunomodulatory potential for application in aquaculture. In tambaqui (Colossoma macropomum), the use of phytogenic additives can strengthen the immune response and reduce damage associated with bacterial infections, such as those caused by Aeromonas hydrophila. Thus, this study evaluated the immunomodulatory effects of dietary supplementation with A. arvensis leaves on histopathological and immunohistochemical parameters (iNOS and CD4⁺) in liver, spleen, and kidney after experimental challenge. One hundred and eight tambaqui were used, distributed into six treatments: negative control (C–), positive control (C⁺), and four groups supplemented with A. arvensis (T1–T4: 10, 20, 30, and 40 g/kg). The fish were fed for 30 days and subsequently challenged with inactivated A. hydrophila (except C–). Liver, spleen, and kidney samples were collected at 12, 24, and 48 hours post-challenge. Histopathological analysis was performed using HE staining and immunohistochemistry for iNOS (innate immunity) and CD4⁺ (adaptive immunity). Quantification was conducted using QuPath, and the normality of residuals and confirmatory statistics were evaluated using GraphPad Prism. Supplementation with A. arvensis promoted dose- and time-dependent immunomodulation. iNOS showed initial peaks (12 hours), indicating early inflammatory activation; T1 and T2 exhibited a more controlled response, while T3 and T4 showed sustained activation without inducing significant lesions. CD4⁺ progressively increased from 24 hours onwards, with higher expression in T3 and T4, evidencing efficient adaptive activation. Histopathology identified lower lesion intensity in the supplemented groups compared to the C⁺ group, which presented congestion, splenic hyperplasia, and renal tubular degeneration. T1 and T2 doses better preserved tissue architecture, while higher doses promoted a robust response without toxicity. No supplemental treatment resulted in necrosis or irreversible damage.

Plants are used for various purposes, including medicinal ones, related to the traditional knowledge accumulated over years by extractive communities. This study aimed to investigate local knowledge about the diversity and use of plants for therapeutic purposes by residents of the Rio Ouro Preto Extractive Reserve – RO. Participants were selected using the snowball sampling method and interviewed using a questionnaire. The length of residence in the reserve ranged from 1 to 74 years. The group that cited the highest number of species was that composed of residents with 1 to 10 years of residence, totaling 42 species and an average of 4.22 species per person. In this research, 51% of the participants were men and 49% women. No significant differences in knowledge were observed according to gender; women cited an average of 4,725 plants, totaling 55 species mentioned, while men cited 68 species, with an average of 4,619 species per person. The vast majority of residents had incomplete primary education (n=48). People with incomplete primary education presented an average of 4.47 species citations per person. Knowledge transmission occurs through various channels, but mainly vertically (56), followed by multiple learning pathways (mixed) (16) and horizontally (5). The least cited forms were individual learning (4) and oblique learning (1). Lamiaceae and Fabaceae were the most cited botanical families. Regarding the plant parts used, aerial structures such as bark, leaves, and stems stand out in various forms of use/preparation, but mainly in the preparation of tea (62.53%), indicated for the treatment of diseases related to the digestive and respiratory systems and general symptoms. The results highlight the importance of medicinal plants for extractive communities, since they are one of the first resources used in the local health system.

The Amazon faces deforestation challenges driven by the expansion of pasturelands, which generates significant environmental impacts and reinforces the need for production models that reconcile environmental restoration and income generation. In this context, agroforestry systems (AFSs) emerge as an alternative by integrating tree and agricultural species within the same production system. Considering the need to expand information on the economic and financial feasibility of these systems, this study analyzes four agroforestry consortia based on açaí palm, established in Porto Velho, Rondônia, Brazil. For the analysis, 1-hectare systems were considered for each arrangement: AFS 1 – açaí, coffee, Brazil nut and cassava; AFS 2 – açaí, banana, cocoa and cassava; AFS 3 – açaí, andiroba and cassava; and AFS 4 – açaí, cupuaçu, banana and cassava. An ex-ante financial analysis was conducted, based on expected results, using actual establishment costs and estimates of post-establishment costs, productivity and prices obtained from the literature, consultation with technical experts and projects with practical experience. The analysis considered a 20-year horizon (2025–2045), with data tabulated in Microsoft Excel spreadsheets. The following indicators were calculated in Excel: Net Present Value (NPV), Internal Rate of Return (IRR), Equivalent Annual Value (EAV), benefit-cost ratio (B/C), payback period and Remuneration of Family Labor (RFL), adopting a discount rate of 6% per year. The results showed that all systems are economically feasible, presenting positive NPV, IRR above the discount rate and benefit-cost ratio greater than one, indicating value generation over time. AFS 1 stood out for its best economic performance, with higher profitability and shorter payback period, while AFSs 2 and 4 showed greater revenue stability due to productive diversification. AFS 3, although feasible, showed lower economic attractiveness. Açaí was consolidated as the main revenue component, exerting strong influence on the financial performance and sensitivity of the systems to price variations. The sensitivity analysis, considering price variations of +10%, +20%, -10% and -20% for the crops, showed that all systems remain feasible even under adverse scenarios, including a 20% reduction in crop prices. Under the models and conditions studied, input costs were found to be concentrated in the establishment phase, while labor demand intensified in the intermediate years, following the increase in management and harvesting activities. RFL proved to be competitive, reaching values three times higher than the regional opportunity cost. Despite their feasibility, the systems showed relatively long payback periods due to low revenue generation in the initial years. Expanding cassava production cycles could help improve cash flow and reduce the payback period; however, this strategy requires adjustments to the spatial arrangement of species, particularly spacing, to avoid excessive competition. It is concluded that the evaluated agroforestry consortia are economically feasible and resilient alternatives for the Amazon. However, the choice of system should consider not only financial indicators, but also factors such as management capacity, labor availability and market access, in order to ensure long-term sustainability.

White-lipped peccaries (Tayassu pecari) and collared peccaries (Dicotyles tajacu) are Neotropical ungulates of high ecological importance, playing key roles in tropical forest dynamics, but have experienced population declines due to forest fragmentation, habitat loss, and anthropogenic pressures, particularly in the Amazon. This study integratively analyzed the influence of environmental and anthropogenic variables on the occurrence, relative abundance, and activity patterns of these species in the Sete de Setembro Indigenous Land (TISS), Rondônia, aiming to support management and conservation strategies in Indigenous territories. A systematic literature review following the PRISMA protocol was conducted to identify key environmental and anthropogenic drivers of T. pecari and D. tajacu occurrence and gaps in existing studies. In parallel, camera trap data from the TISS were analyzed to estimate relative abundance and characterize daily activity patterns and temporal overlap using independent records and circular statistics. Environmental and land-use variables derived from remote sensing, including vegetation structure and landscape metrics, were evaluated at multiple spatial scales using quantitative models. The results showed a higher number of independent records for D. tajacu, suggesting greater relative occurrence or detectability, while T. pecari was less frequently recorded. Both species were predominantly diurnal, with D. tajacu showing a clearly diurnal pattern and T. pecari a cathemeral pattern with greater temporal concentration of activity. Despite high temporal overlap, differences in daily time use were evident. The literature synthesis and multiscale analyses indicated that forest integrity, resource availability, and anthropogenic pressure strongly influence species occurrence, with T. pecari being more sensitive to habitat loss and fragmentation and D. tajacu more tolerant of heterogeneous landscapes. Integrating temporal, spatial, and environmental information enhances understanding of the ecology of these species and provides critical insights for their conservation in Amazonian Indigenous territories.
