factors of Candida spp. isolates causes of infections in patients in the region of
Porto Velho, RO, Brazil.

SUMMARY:

The Candida genus yeasts colonize the human organism in a commensal form, only,
depending of the state of the host, these microorganisms can to comport as
opportunistic agents and cause from superficial infections to systemic infections.
These infections result in prolonged hospitalizations, increased hospital expenses, in
besides high numbers of morbidity and mortality. Therefore, the objective of this study
was to characterize, at the species level, clinical isolates of Candida yeasts from
patients in the Porto Velho, Rondônia region, as well as to phenotypically determine
the virulence and susceptibility to antifungal agents. For this, the specimens were
obtained from patients attended at the Medical Mycology laboratory of the Research
Center in Tropical Medicine of Rondônia (CEPEM/RO), that went isolated and
characterized primarily through conventional of the identification methods. The isolates
were sequenced using the ABI 3100 Prism sequencer, series 1592-015 (Applied
Biosystems) for molecular identification of the species. The disk diffusion method was
used coursing the recommendations of the M44-P protocol to verify the susceptibility
of the isolates to antifungal agents. For phenotypic determination of hemolysin and
phospholipase activity, test on egg yolk agar and sheep of the blood agar were used.
Through the methodologies used, the present study evidenced the presence of three
Candida species as causes of infections in patients in the region of Porto Velho,
Rondônia, of which C. albicans (78.9%) was the most prevalent, followed by C.
tropicalis (15.8%) and C. parapsilosis (5.3%). We also determined high numbers of
resistance to fluconazole (89.5%) and itraconazole (78.9%), evidencing a reduction in
the sensitivity of Candida spp. to first option triazole antifungals. Besides, it was
demonstrated that 100% of Candida isolates were hemolysin producers and that only
C. albicans were phospholipase producers, corresponding to 52.4% of the isolates of
the species. These results provide important information for the management of
infections in relation to strategies of the treatment, control and prevention of the cases
of candidiasis diagnosed in the region.
