SUMMARY:

In 2020, nearly half of the world's population was at risk of contracting malaria. In the same year, there were about 241 million cases of the disease and 627,000 deaths worldwide. In Brazil, about 99% of malaria transmission occurs in the Legal Amazon and P. vivax is the predominant parasite in the Americas region, where it is the cause of 75% of malaria cases. The clinical sign of malaria can range from mild manifestations to severe and lethal conditions, with the most common symptoms being intense physical weakness, fever, nausea, headache, myalgia and vomiting. Adults and children can develop severe and complicated malaria, which can be fatal in some cases. In vivo and in vitro studies have shown that the p53 protein and the cytokine IFN-γ may be involved in protective mechanisms against Plasmodium infection. The objective of this study was to analyze the level of expression of the TP53 and IFNG genes in the infection caused by P. vivax in different parasitemias in patients assisted at the Centro de Pesquisa em Medicina Tropical – CEPEM. A total of 53 samples were collected, of which 24 were randomly selected for a previous study, being classified into 4 groups with different parasitemia +/- (half cross), + (a cross), ++ (two crosses) and +++ (three crosses). Extraction and purification of total RNA from whole blood and cDNA synthesis were performed. The ideal primer concentration and cDNA efficiency were performed to standardize the real-time PCR technique. To evaluate the variation in expression, the comparative Cq methodology was used, given by the ΔCT equation: Cq gene of interest – Cq of the control gene. Cq values of the TP53 and IFNG genes were obtained, using the GAPDH gene as an endogenous control. It was observed that patients with high parasitemia +++ (three crosses) had a lower expression profile of the TP53 gene compared to patients with lower parasitemia. The IFNG gene showed lower expression in patients with less parasitemia +/- (half cross) and higher expression in patients with ++ (two crosses) parasitemia. The results corroborate studies carried out in vitro and in vivo that p53 may be involved in mechanisms of resistance to Plasmodium
