This research aimed to develop topical biocosmetic formulations with antioxidant properties from the ethanolic extract of the bulbs of Eleutherine bulbosa (Mill.) Urb. (Iridaceae), a medicinal plant native to the Amazon, popularly known as marupazinho. The bulbs were collected on the campus of the Federal University of Rondônia Foundation, in Porto Velho-RO, identified (voucher RB 371008) and subjected to controlled drying (45 °C) and extraction by cold maceration with 96% ethanol for 21 days, obtaining a yield of 2.15% (m/m). Phytochemical characterization was performed by ultra-high performance liquid chromatography coupled to high-resolution mass spectrometry (UHPLC-HRMS) in positive and negative modes, identifying classes of secondary metabolites such as flavonoids, naphthoquinones (eleutherin, isoeleutherin, eleutherol), anthraquinones, phenolic acids, xanthones, terpenoids, and glycosides. In vitro antioxidant activity by the DPPH method revealed an IC50 of 89.85 µg.mL-1, classified as strong. The hemolysis assay in human O+ erythrocytes demonstrated low toxicity, with hemolysis less than 6% at a concentration of 100 µg.mL-1, indicating safety for topical use. Three cosmetic bases (gel, cream, and gel-cream) were developed incorporating the ethanolic extract at concentrations of 0.02%, 0.2%, 0.5%, 1.0%, and 2.0% (w/w), totaling 45 formulations. The organoleptic characteristics, pH (5.5–6.5), spreadability, and homogeneity met dermocosmetic standards. The accelerated stability study (60 days; 15–25 °C and 35–45 °C) showed maintenance of properties in most formulations, with minimal changes only at the highest concentrations in cream and gel-cream. The results demonstrate that the ethanolic extract of E. bulbosa presents a rich phytochemical profile, high antioxidant capacity, low hemolytic cytotoxicity, and good compatibility with conventional cosmetic vehicles, configuring itself as a promising natural active ingredient for the prevention of cutaneous oxidative stress and premature aging.
