Natural products from plants are important and promising sources of chemical compounds, so that they can be transformed into medicines. Eleutherine bulbosa is a member of the Iridaceae family, present in South America. Eleutherine bulbosa is used in menstrual and intestinal disorders, as an abortive and contraceptive agent. From this information, there was an interest in studying this plant that has great phytotherapic potential. This work aimed to evaluate the leishmanicidal potential of the ethanolic extract and CHCl₃, EtOAc and acetone fractions. The test for class of substances was carried out that revealed the presence of flavonoids, alkaloids, terpenes and quinones. The effect of ethanolic extract and fractions was evaluated in relation to the promastigote forms of L. amazonenses and L. infantum, and cytotoxicity against J774 cells and the selectivity index was calculated. For the ethanolic extract, growth inhibition of promastigote forms was observed in the highest concentration tested (200µg.ml^-1), 28% for L. amazonenses and 70% for L. infantum. For the CHCl₃ fraction, 99% inhibition for L. amazonenses and 100% for L. infantum in the highest concentration tested (200 µg.ml^-1). Regarding the EtOAc and acetone fractions, no inhibition was observed at the highest concentration tested. The ethanolic extract was active for the species L. infantum (IC₅₀ of 51.6 µg.ml^-1). Of the three fractions tested, only CHCl₃ was active for both species, with IC₅₀ values of 50.1 µg.ml^-1 for L. amazonensis and 21.85 µg.ml^-1 for L. infantum. Regarding the selectivity index, the ethanolic extract presented a value of 0.36 for L infantum and the fraction CHCl₃, 0.62 and 1.44, for L. amazonensis and L. infantum respectively.
