BACTERICIDAL AND LEISMANICIDAL ACTIVITY OF Protium calendulinum Daly AND Protium robustum (Swart) D. M. Porter

The genus Protium is widely distributed in South America, with the Amazon region being the center of its biodiversity. Various parts of its plants are used in traditional medicine to treat different types of illnesses, and its resin is used in religious ceremonies and as an insect repellent. The species Protium calendulinum and Protium robustum, found mainly in the Northern Region of Brazil, had their microbiological activities evaluated using the serial microdilution method. To verify bactericidal activity, ethanolic extracts of P. calendulinum and P. robustum, and fractions of P. calendulinum, CHCl3, EtOAc, acetone, and MeOH were used against the species Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Acinetobacter baumannii. The essential oil of P. calendulinum breu was evaluated for its bactericidal activity using the disk diffusion method against Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Acinetobacter baumannii, and Enterococcus faecalis. For the leishmancidal activity of P. calendulinum against Leishmania amazonensis, its ethanolic extract, hexane fractions, CHCl3, EtOAc, acetone and MeOH, breu, and breu essential oil were tested; for Protium robustum, its ethanolic extract was tested. Concomitantly with the biological tests, chemical composition analyses of the essential oil of P. calendulinum resin and essential oils from green and ripe P. robustum fruits were performed by GC-MS. Promising bactericidal activity was observed for the methanolic fraction of P. calendulinum against A. baumannii with a MIC of 97.75 µg·mL-1 in a serial microdilution test. For the essential oil of P. calendulinum resin, in a disk diffusion test, inhibition halos were formed against S. aureus (10 mm), MRSA (7.7 mm); P. aeruginosa (7.7 mm); K. pneumoniae (8.3 mm); and E. faecalis (14.0 mm). The essential oil of breu, at a concentration of 100 µg·mL-1, against L. amazonensis, showed a Growth Inhibition Index (GII) of 93%, and at a concentration of 200 µg·mL-1, a GII of 100%, the observed MIC was 100 µg·mL-1 and IC50 < 200 µg·mL-1. The breu of P. calendulinum, dissolved in DMSO, demonstrated leishmanicidal activity with an MIC of 100 µg·mL-1 and IC50 < 200 µg·mL-1. Analysis of the chemical composition of the essential oil of breu indicated ortho-cymene (27.52%), α-phellandrene (17.73%), α-pinene (13.77%), eucalyptol (6.47%), and α-terpinene (5.97%) as the major compounds, while analysis of the essential oils of green and ripe fruits of P. robustum showed bicyclogermacrene (23.17%), germacrene D (11.11%), limonene (7.09%), α-copaene (6.87%), and α-pinene (5.84%) as the major compounds. Based on the observed results, it is believed that studies with Protium calendulinum and P. robustum should continue in order to obtain a possible source of new drugs, or to indicate their oils and breu as objects of future studies in the fight against microorganisms of clinical importance.
